SAMPLE: : 400 seeds

PROCEDURE:

1. Randomly select 400 seeds; 4 replicates of 100. Place into 125 ml flasks.

2. Add 30 ml of sterile DI water.

3. Shake for 15 minutes.

4. Pour solution into centrifuge tubes.

5. Centrifuge at 3000 rpm for 15 minutes.

6. Pour off remaining suspension and retain pellet.

7. Examine resuspended pellet for urediniospores or teliospores of P. pachyrhizi under a compound microscope.
